Life or humanity.
Life is a mystery as to how it was created, as was how it was perfected (Or if it even is perfected yet), and the rest... I dunno.
Humanity because the origins of humanity are still debated, as are the ways it was shaped (ID or evolution) and humans created fire. They die too.
It'd suck if I'm wrong on both counts.

I'd say everything and God, because "its creation" refers to the universe, "he" refers to God, and the rest of it speaks of how the world rests on his consience (should he have one), and death would "knock at his door" if the whole universe is destroyed and make his "heart ring" with sorrow.

Only other thing I can think of is fire or some kind of engulfing flame.
What mysteries are in its creation?
Who's hand did bend its ore?
Where did the knowledge come from?
And could he have made any more?
On his finger it did lie,
Yet on his soul the more.
For the fire it would bring,
Would make his heart ring,
And death, would come knocking at his door.

I'm gonna say Fire. Or something to do with fire.
No one truly knows who first created fire (More or less discovered) which explains the first four lines. Who modified it, who discovered, blah blah blah.
The finger... Ehhhh, I guess I could say the "Him" they're referring to is humanity, so it being on the finger tip is that the technology was always availible for use (Cut me some slack, no one else is trying). I don't really know how to explain the two before the last, but fire kills, so remember:

Would make his heart ring, -I'm guessing a flame randomly popping up outta nowhere is a bit scary
And death, would come knocking at his door. -You catch on fire, you die.
WAIT! HAHAH! WOOO! I just watch LoTR. The ring of POWA

The forest, or trees.
They feed people with their fruit. Houses are built with wood from trees. Paper's made from trees, and so are pencils (Mostly). But when there's no more forests left, there ya go.
Oh, and they fertilize the ground when they die. And when they live.
