Not always. My mom's, for instance, has onboard video and three PCI slots. No AGP at all.
Fortunately, even if you're stuck with PCI, you'll be able to find a card and get playing

Still, if you've got a graphics accelerator running off the motherboard instead of an actual video card, the others are right that there should be slots of some kind. If you have the documentation for your motherboard around somewhere, that'd be an easy way to find out. You're probably looking for AGP or PCI slots, I doubt you'd have any PCI-e. If the most you want out of your computer is to be able to play Guild Wars, then it shouldn't be too difficult to pick up a pretty cheap card that would do that just fine, the specs aren't all that demanding.
