Nature Runes... Hmm... Killing giants, hobgoblins, or darkwizards is a decent way of getting them, and Nature Rune isle in the NE corner of the wilderness(next to lessers) has 4 in one pile and 3 in the other, but you need to telegrab them. As for a safe respawn, I can't remember because I haven't played in along time(though I don't think there is one). Sorry.

natures are dropped by ice warriors 3 at a time, dark wizards drop them but only 1 at a time, ice giants drop them but not as often as the warriors, also moss giants cant think of any others

normal giant drop 4 at a time
